Principle:

Bile Salt Agar is formulated as per recommended by BIS (1986) for isolation, identification and enumeration of Vibrio cholerae. The media is composed of peptone, special peptone, sodium chloride, sodium taurocholate and agar. The bile salt provides selectivity for the growth of Vibrio species, and many other gram-negative bacteria. On bile salt agar colonies of Vibrio's have a distinctive appearance and can be easily differentiated from Escherichia coli. The medium contains peptone, special peptone (equivalence to meat extract) which provides carbonaceous, nitrogenous compounds and other essential growth nutrients. Sodium chloride maintains osmotic equilibrium of the medium. Sodium taurocholate inhibits most of the gram negative organisms.

Use:

Recommended by BIS committee under the specifications IS:5887 (Part V) -1976 for isolation and identification of bile tolerant bacteria responsible for food poisoning.

Directions:

Dissolve 40.00 grams in 1000 ml distilled water. Heat to boiling to dissolve the medium

completely. Sterilize by autoclaving at 15 lbs pressure (121°C), cool to 45-50°C. Mix well and distribute aseptically in petri plates and allow to solidify. Ensure complete solidification and inoculate test sample aseptically.

 

Storage and Shelf Life:

The product is highly hygroscopic; keep the container tightly closed at all times and store it properly as per the conditions mentioned on the label. The declared expiry is valid only when stored as per the conditions mentioned on the label.

Note:

Sterilize media immediately after reconstitution.

Disposal:

To avoid the contamination or propagation of any hazardous microbes the used, unusable or modified preparation of this product must be disposed after autoclaving after completion of task.
