Chocolate Agar Base, composed of proteose peptone that provides the organism with nitrogenous nutrients, amino acids, and other elements essential for the growth of the organisms. Dextrose is carbon and energy source. Phosphates buffer the medium. Sodium chloride maintains osmotic balance. Agar is a solidifying agent. The selectivity is provided by addition of glutamine, coenzymes, carboxylase, hematin and growth factors required by fastidious organisms. Thayer-Martin Media is a chocolate agar supplemented with vancomycin, nystatin, and colistin to inhibit the normal flora, including nonpathogenic Neisseria for the selective isolation of N. gonorrhoeae and N. meningitidis.
Use: Recommended for the isolation of Neisseria gonorrhoeae from chronic and acute cases of gonococcal infections. |

Directions: Dissolve 45.50 grams in 500 ml distilled water. Boil to dissolve the medium completely and sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min, cool it to 42-45 °C. Add 500 ml of sterile hemoglobin solution and mix thoroughly and distribute aseptically in petri plates. Ensure complete solidification and inoculate test sample aseptically.
Sterile 2% hemoglobin solution: Add 10 ml of hemoglobin to distilled water and bring volume to 500 ml. Mix thoroughly and autoclave for 15 min at 15lbs pressure at 121°C. Cool at 45°C -50°C.
The media can also be enriched with yeast autolysate supplement or vitamin growth supplement reconstituted. Mix well before pouring.
Storage and Shelf Life: The product is highly hygroscopic; keep the container tightly closed at all times and store it properly as per the conditions mentioned on the label. The declared expiry is valid only when stored as per the conditions mentioned on the label.
Note: Sterilize media immediately after reconstitution.
Disposal: To avoid the contamination or propagation of any hazardous microbes the used, unusable or modified preparation of this product must be disposed after autoclaving after completion of task.
